To test whether aveR affects the transcription of oligomycin biosynthetic genes, semiquantitative RT-PCR analyses were performed to detect the expression of two selected polyketide synthase (PKS) genes (olmA1 and olmA4) and two putative pathway-specific regulatory genes (olmRI and olmRII) in olm cluster. All the detected olm genes were upregulated in the aveR mutants. In ChIP assays, the predicted promoter regions of the olm genes were amplified from both positive control DNA and anti-AveR-immunoprecipitated DNA. |
